He emphasized that to avoid non-specific staining caused by residual reagents such as primary and secondary antibodies, thorough washing is essential. It's important to increase both the time and frequency of washing. Typically, it's recommended to wash three times for three minutes before incubation with the primary antibody, and five times for five minutes after the primary antibody incubation.
**Precautions:**
1. **Wash separately** to prevent contamination from cross-reactions.
2. **Gently rinse** to avoid peeling off the tissue sections. The dipping method is recommended.
3. **Ensure sufficient rinsing time** to completely remove any bound substances.
4. **Use PBS with the right pH and ionic strength**. It is advised to use 0.01 M phosphate-buffered saline (PBS) at a pH of 7.4–7.6. Neutral or slightly alkaline conditions (pH 7–8) are beneficial for immune complex formation, while acidic conditions can lead to decomposition. Low ionic strength favors immune complex formation, while high ionic strength promotes dissociation.
